Histamine-induced and chloroquine-induced intracellular calcium increases were reduced in TLR4 KO neurons. A and B. DRG neurons loaded with Fura2-AM were treated with histamine (10 μg/ml) and chloroquine (CQ, 10 μg/ml) and intracellular calcium level was monitored by calcium imaging. Percentage of responding cells (A) and average net increase of [Ca++]i (Δ ratio (340 nm/380 nm)) in the responding population (B) are in graphs (n = 5, 544 cells measured; *p < 0.005).
