LRRK2 induces p21 transcription in differentiated SH-SY5Y cells. Differentiated SH-SY5Y cells were transfected with the shLRRK2 plasmid for 18 h (a) or the myc-tagged G2019S for 24 h (b). cDNA was synthesized from mRNA isolated from the cell lysates in each case. Semi-quantitative PCR was carried out using the cDNA as template, with the specific primers (Table 1) for the indicated genes. All experiments were repeated at least three times, and a representative result is shown with the statistical analysis. c. Activity of the p21 promoter-luciferase reporter from cells transfected with shLRRK2, G2019S mutant, or p53 WT and T304/377D.
